Introduction About LAREGAB AT LS 100/10MG TABLET

LAREGAB AT LS 100/10MG TABLET is a combination of Amitriptyline and Gabapentin which belongs to the group of medicines called Tricyclic Antidepressants and Antiepileptics. It is used in the management of neuropathic pain in adults. Neuropathic pain is a long-term weakness, numbness or pain caused by damage to the nerves in hands and legs characterized by a feeling of hot, burning, throbbing, shooting, sharp, tingling, numbness.

LAREGAB AT LS 100/10MG TABLET is not recommended for use in patients with severe liver disease and heart problems and it should be used with caution in patients having kidney problems or undergoing haemodialysis. Do not stop taking LAREGAB AT LS 100/10MG TABLET suddenly, as this may lead to increased risk of seizure.  

LAREGAB AT LS 100/10MG TABLET is not recommended for use in pregnant woman especially during the last few months of the pregnancy. Do not take LAREGAB AT LS 100/10MG TABLET if you are breast-feeding as it may pass through breast milk to the baby. Consult your doctor for advice before taking it.   

LAREGAB AT LS 100/10MG TABLET is not recommended for use in children and adolescents below 18 years of age and should be used with caution in elderly patients aged 65 years and above. Consult your doctor for advice before taking LAREGAB AT LS 100/10MG TABLET.

The most common side effects of taking LAREGAB AT LS 100/10MG TABLET are dizziness, drowsiness, muscle pain, nausea, vomiting and stomach pain. Consult your doctor if any of these side effect worsens.

Uses Of LAREGAB AT LS 100/10MG TABLET

  • Manage neuropathic pain (long term pain caused by damage to the nerves) in adults

How LAREGAB AT LS 100/10MG TABLET Works

LAREGAB AT LS 100/10MG TABLET works effectively in reducing the neuropathic pain where Amitriptyline effects are related to its mood-altering activities which acts by increasing the concentration of the neurotransmitters called serotonin and norepinephrine as these substances are found to be low in the anxiety patients and Gabapentin acts by blocking the pain signals travelling through the damaged nerves and the brain and hence manages nerve pain.

How to use LAREGAB AT LS 100/10MG TABLET

Take LAREGAB AT LS 100/10MG TABLET as advised by your physician. Swallow LAREGAB AT LS 100/10MG TABLET with a glass of water. Do not crush or chew the medicine. Your doctor will decide the correct dose and duration of therapy for you depending upon your age, body weight and health condition. Do not stop taking LAREGAB AT LS 100/10MG TABLET without consulting your doctor.

Side Effects Of LAREGAB AT LS 100/10MG TABLET

Common

  • sleepiness/drowsiness, dizziness, lack of coordination, tiredness
  • dizziness while standing up (orthostatic hypotension)
  • irregular or fast heartbeat, heart block
  • eating disorder characterized by low body weight, increased appetite
  • anger, nervousness, difficulty with thinking, confusion, mood changes, depression, anxiety
  • jerky movements, loss of memory, tremor, headache, sensitive skin, decreased sensation (numbness), unusual eye movement, increased, decreased or absent reflexes, difficulty with speaking and coordination, difficulty sleeping
  • blurred vision, double vision
  • feeling of spinning (vertigo)
  • high blood pressure
  • difficulty in breathing, sore throat, cough, dry and congested nose
  • vomiting, nausea
  • problems with teeth, inflamed gums
  • stomach pain, diarrhea, indigestion, constipation, flatulence, dry mouth or throat
  • facial swelling, bruises, rash, itching, acne
  • muscles and joint pain, back pain, muscle twitching
  • difficulties with erection (impotence) in male
  • swelling in the legs and arms, weakness, pain, feeling unwell, difficulty with walking
  • increase in weight
  • rhythmic shaking movements of body parts
  • excessive sweating (anxiety)
  • slurred or slow speech
  • agitation (anxiety)
  • sexual disturbances
  • disturbance in attention
  • altered taste sensation
  • dilated pupils

Uncommon

  • allergic reaction such as hives
  • decreased movements
  • difficulty in swallowing
  • swelling of the face, limbs and trunk
  • excitement, nightmares
  • fits, ringing in ears
  • nettle rash (urticaria)
  • difficulties passing urine
  • increased breast milk production or its outflow without breast feeding
  • increased pressure in the eyeball

Rare

  • loss of consciousness
  • decreased appetite
  • delirium (especially in elderly patients), hallucinations
  • swelling of the salivary glands
  • hair loss
  • increased sensitivity to sunlight
  • breast enlargement in men
  • weight loss

Stop taking LAREGAB AT LS 100/10MG TABLET and contact your doctor immediately if you experience any of the following side effects:

  • swelling of the skin, lips and face
  • rash and redness and/or hair loss
  • signs of acute pancreatitis such as nausea and vomiting, persistent stomach pain
  • breathing problems, swollen glands 
  • severe weakness, unusual bruising or bleeding
  • frequent infections, muscle pain
  • hives (nettle rash), facial swelling due to allergies (Quincke’s oedema or angioedema), difficulty in breathing, severe dizziness with very fast heartbeat and sweating
  • blistering of the skin and rapid deterioration of general condition, peeling of skin (including slight bleeding) of eyes, nose, mouth/ lips or genitals (Stevens Johnson-Syndrome, Lyell-Syndrome, Erythema multiforme) and sensitivity to light
  • eye pain, intermittent blurred vision, rainbow vision
  • jaundice (yellowing of the skin or white of the eyes)
  • persistent sore throat and fever
  • suicidal thoughts

How To Manage Side Effects

Nausea or Vomiting:

Take LAREGAB AT LS 100/10MG TABLET with or just after meals. Stick to simple meals. Try to avoid oil rich or spicy foods, sugary snacks and drinks. Try to drink liquid slowly along with your meals or in between meals. Consult your doctor if the symptom worsens.

Dizziness:

Lie or sit down immediately when you feel dizzy. If you’re feeling severe episode of spinning (vertigo), lie in a dark and quite place with eyes closed. Try to avoid vehicle driving. Discuss with your doctor if the symptom worsens.

Headache:

Apply hot or cold-water bag on your head. Try to take rest in a quiet and dark room. Drink tea or coffee. Massage on scalp or head with mild pressure. Consult your doctor if the symptom worsens.     

Dry mouth:

Dry mouth can be managed by chewing sugar-free gum or sucking on sugar-free hard candies to stimulate the flow of saliva. Sip water at regular intervals of time. Do not use mouthwashes that contain alcohol because they can be drying. Consult your doctor if the symptom worsens.     

Stomach pain:

Try to rest and relax. Try to have smaller and frequent meals. Keep a heat pad on your stomach. Talk to your if the symptoms does not improve.

Warning & Precautions

Pregnancy

LAREGAB AT LS 100/10MG TABLET is not recommended for use in pregnant woman unless considered clearly necessary, especially during the last few months of pregnancy as the new-born may have withdrawal symptoms such as tremors (shaking movements), irregular breathing, irritability, increased muscle tension, poor drinking, crying loudly, urinary retention and constipation. Consult your doctor before taking LAREGAB AT LS 100/10MG TABLET.

Breastfeeding

Do not take LAREGAB AT LS 100/10MG TABLET if you are breast-feeding as it may pass through breast milk to the baby. Consult your doctor before taking LAREGAB AT LS 100/10MG TABLET.

Driving and Using Machines

Do not drive or operate any machines because LAREGAB AT LS 100/10MG TABLET may cause dizziness, drowsiness and tiredness.

Alcohol

Avoid consumption of alcohol while taking LAREGAB AT LS 100/10MG TABLET as it may cause sleepiness or dizziness.

Kidney

LAREGAB AT LS 100/10MG TABLET should be used with caution in patients with kidney problems or those who are undergoing haemodialysis. Consult your doctor before taking LAREGAB AT LS 100/10MG TABLET.

Liver

LAREGAB AT LS 100/10MG TABLET is not recommended for use in patients having severe liver disease. Consult your doctor before taking LAREGAB AT LS 100/10MG TABLET.

Allergy

Do not take LAREGAB AT LS 100/10MG TABLET if you are allergic to Amitriptyline or Gabapentin.

Lungs

LAREGAB AT LS 100/10MG TABLET should be used with caution in patients with respiratory disorders. Consult your doctor before taking LAREGAB AT LS 100/10MG TABLET.

Heart Disease

LAREGAB AT LS 100/10MG TABLET is not recommended for use in patients who had heart attack (myocardial infarction), disturbances in heart rhythm, heart block or coronary artery disease. It should be used with caution in patients with pre-existing heart disease. Consult your doctor before taking LAREGAB AT LS 100/10MG TABLET.

Others

Before taking LAREGAB AT LS 100/10MG TABLET, inform your doctor if you:

  • have acute pancreatitis with signs of persistent stomach pain, nausea and vomiting
  • have nervous system disorders
  • have a problem resulting in low levels of potassium or magnesium or high levels of potassium in the blood
  • have a planned surgery
  • have an overactive thyroid gland

Use in pediatrics:

LAREGAB AT LS 100/10MG TABLET is not recommended for use in children and adolescents below 18 years of age. Consult your child’s doctor for advice before administration of LAREGAB AT LS 100/10MG TABLET.

Use in geriatrics:

LAREGAB AT LS 100/10MG TABLET should be taken with caution in elderly patients (aged 65 years or above) especially in patients with impaired kidney function. Consult your doctor before taking LAREGAB AT LS 100/10MG TABLET.

Interactions

A. Drug – Drug interactions:

Before taking LAREGAB AT LS 100/10MG TABLET, inform your doctor, if you are taking any of the following medicines:

  • antacids such as aluminum hydroxide and magnesium hydroxide (medicines used to reduce heart burn, indigestion)
  • fluoxetine, paroxetine, fluvoxamine, duloxetine, bupropion and monoamine oxidase inhibitors like phenelzine, iproniazid, isocarboxazid, nialamide or tranylcypromine (medicines used to manage depression)
  • terbinafine, itraconazole, fluconazole, ketoconazole, (used to manage fungal infections)
  • erlotinib (medicine used for certain types of cancer)
  • warfarin and phenprocoumon (medicines used as a blood thinning agent)
  • rifampicin (medicine used to manage tuberculosis)
  • St john’s wort (hypericum perforatum) (used to manage mild depression)
  • glyceryl trinitrate, adrenaline, ephedrine, isoprenaline, noradrenaline, atenolol, propranolol (medicines used to manage heart disease)
  • phenylephrine and phenylpropanolamine (medicines used to manage cough and cold)
  • medicines to manage high blood pressure (Ex. diltiazem and verapamil, betanidine, reserpine, methyldopa, guanethidine and clonidine)
  • medicines to manage parkinson’s disease and gastrointestinal disorders (Ex. atropine, hyoscyamine)
  • thioridazine (medicine used to manage schizophrenia)
  • nefopam, tramadol, morphine, buprenorphine (used to manage pain)
  • barbiturates like phenobarbital, pentobarbital (medicines used to manage insomnia) or other sleeping medicines like triazolam, zolpidem
  • cimetidine (used to manage stomach ulcers)
  • methylphenidate (used to manage ADHD-attention-deficit/hyperactivity disorder)
  • ritonavir (medicine used to manage HIV)
  • oral contraceptives
  • valproic acid, phenytoin and carbamazepine (medicines used to manage epilepsy)
  • thyroxine (used to manage underactive thyroid)
  • baclofen (medicine used as muscle relaxant)

Overdosage:

If you or anyone else accidentally takes too much of LAREGAB AT LS 100/10MG TABLET, consult your doctor immediately or visit the nearby hospital. Overdosage symptoms of LAREGAB AT LS 100/10MG TABLET includes loss of consciousness, dizziness, double vision, slurred speech, drowsiness, diarrhoea, dilated pupils, fast or irregular heartbeats, difficulties passing water, dry mouth and tongue, intestinal blockage, low blood pressure, weak pulse, pallor (pale skin), difficulty in breathing, blue discolouration of the skin, decreased heart rate, drowsiness, loss of consciousness, coma, fits, fever, agitation, confusion, hallucinations, uncontrolled movements, various cardiac symptoms such as heart block, heart failure, cardiogenic shock, metabolic acidosis, low levels of blood potassium (hypokalaemia).
